As we journey through life, it’s natural to notice changes in our physical strength and stamina. The gradual loss of muscle mass, strength, and function associated with aging, a condition known as sarcopenia, affects a significant portion of adults over 50 [2]. While resistance training and protein intake remain the cornerstones of muscle preservation, researchers are exploring new avenues that target the very building blocks of our cells.
One of the most intriguing areas of longevity science involves our mitochondria. These are the tiny powerhouses inside our cells, responsible for generating over 90% of the energy we need for life and movement. With age, mitochondrial function can decline, impacting everything from energy levels to muscle performance. This has led scientists to investigate compounds that could help support mitochondrial health, with a particular focus on a molecule called Urolithin A.
What the Research Shows
Urolithin A is not something you eat directly. Instead, it is a postbiotic compound, meaning it’s produced by our gut bacteria when they digest specific precursors called ellagitannins and ellagic acid. These precursors are abundant in foods like pomegranates, raspberries, strawberries, and walnuts [4].
Initial interest in Urolithin A was sparked by pre-clinical research. In a foundational 2016 study, scientists observed that Urolithin A could trigger a cellular recycling process called “mitophagy,” which clears out old, dysfunctional mitochondria to make way for new ones. In this early work, Urolithin A was shown to extend the lifespan of C. elegans worms and improve muscle function in rodent models [3].
While promising, these findings required validation in human studies. A significant step forward came from a 2022 randomized clinical trial published in JAMA Network Open. In this study, researchers investigated the effects of Urolithin A supplementation in 88 healthy older adults between the ages of 65 and 80 [1].
Over four months, participants were given either a daily dose of 1,000 mg of Urolithin A or a placebo. The results were noteworthy. The group receiving Urolithin A showed a significant improvement in muscle endurance, measured by the number of repetitions they could perform during a hamstring exercise, compared to the placebo group [1]. Researchers also noted that levels of several biomarkers linked to mitochondrial health and reduced inflammation were improved in the blood of those taking the supplement [1].
It is important to report the findings with nuance. While endurance improved, the study did not find statistically significant improvements in the six-minute walk test or in maximal ATP production in the hand muscle [1]. This suggests Urolithin A may have specific effects on sustained muscle performance rather than overall strength, a topic that requires further investigation.
What This Means for Everyday Wellness
This research is an exciting development in the science of healthy aging, highlighting a potential link between a gut-derived compound, cellular energy, and physical endurance. It underscores a shift in wellness from just focusing on muscles to supporting them at a microscopic level.
One crucial takeaway is the variability in our ability to produce Urolithin A. The production of this compound depends entirely on having the right balance of gut microbes, and studies suggest that not everyone is an efficient producer. This may explain the growing interest in direct supplementation.
However, this research does not diminish the foundational importance of a healthy lifestyle. The most evidence-based strategies for combating age-related muscle decline remain consistent and accessible:
- Regular Physical Activity: A combination of aerobic exercise and resistance training is critical. For older adults, this includes at least 150 minutes of moderate-intensity activity a week and muscle-strengthening activities on 2 or more days a week [5].
- Adequate Protein Intake: Consuming sufficient high-quality protein helps provide the building blocks for muscle repair and synthesis [2].
- A Nutrient-Rich Diet: Including foods rich in ellagitannins, like berries and nuts, can provide your gut microbiome with the precursors to potentially make Urolithin A, alongside countless other benefits from a balanced diet [4].
A Gentle Reminder
The science of cellular aging is moving quickly, offering new insights into how we can support our bodies as we get older. Before considering any new supplement, including Urolithin A, it is essential to consult with your healthcare provider to discuss whether it is appropriate for your individual health circumstances and goals.
